Oddly enough, this is not someone who skins mules. It was a man and mule (the man would generally own the mule) who would be hired (as a team) as day labor by the railroads. The work generally involved moving very heavy things. Jimmie' Rodgers song "Muleskinner Blues", later covered by Bill Monroe brought the term into mainstream American culture.
